The Effect of Estradiol-17Β Levels in Estrous Cow Sera on In vitro Maturation and Fertilization of Cow Oocytes

- The aim of this study was to compare influence of Estradiol-17Β levels measured in estrous cow sera (ECS) on oocytes from slaughtered in vitro maturation and fertilization with fetal calf sera (FCS). Three Holstein Fresian heifers were used for source of the sera. Blood was collected in the first 3 hours of the estrous from heifers and sera was decomposed from the blood then sera were mixed and stock serum was obtained. Level of estradiol-17Β in all the sera was analized by Radio Immuno Assay. Oocytes were selected and used as A and B quality grade morphologically. Stock serum was added in the maturation media 5%, 10% and 20%. The same proportions of FCS were added in the control group. Data were tested by ANOVA. Estradiol-17Β (E2) levels in the heifers blood sample were found for Heifer1, Heifer2 and Heifer3 and stock serum as respectively; 18.4 pg/ml, 20.1 pg/ml, 21.8 pg/ml and 19.9 pg/ml. Estradiol 17-Β was also measured 34.8 pg/ml in the FCS. For adding 5%, 10% and 20% proportions of ECS stock serum, the results were respectively; 0.78±0.010a, 0.84±0.009b, 0.86±0.012b maturation (p<0.01). The results of addition of FCS as a control group for 5%, 10% and 20% sera levels were 0.77±0.010a, 0.82±0.008b, 0.85±0.010b maturation (p<0.01); 0.51±0.020a, 0.51±0.023a, 0.59±0.017b fertilization result (p<0.01).

As a result, estrous cow serum can be used economically for in vitro cow oocytes maturation. When ECS and FCS 10% and 20% were added in the maturation media, more better maturation and fertilization results were found according to 5% ECS and FCS.
