Effect of Some Cryoprotectans in Cryopreservation of Angora Buck Semen

- The aim of this study was to investigate the effects of some cryoprotectants added to extender of Angora buck semen on spermatologic parameters after frozen-thawed semen. Ejaculate samples obtained from three mature Angora bucks were used in this study. Ejaculates were collected using an artificial vagina twice a week, for five weeks running, during the reproductive season. After pooling, each pooled ejaculate was split into four equal aliquots and diluted with based extender containing cryoprotectants [100 mM trehalose + 0.015 g/10 ml EDTA, 5 mM GSSG and trehalose + EDTA + GSSG (combined)], or no cryoprotectants (control) at 37oC. Diluted samples were aspirated into 0.25 ml straws, equilibrated at 5oC. Following equilibration, the straws were frozen in liquid nitrogen vapour and stored at -196oC so as to be evaluated.

Post-thawed spermatozoa motility did not exist significant difference at the groups with GSSG and combined (53.7±3.7%, 46.2±1.2%), compared to the control (42.5±3.2%). But groups including trehalose+EDTA significantly deteriorated the spermatozoa motility, compared to the other groups. At abnornal spermatozoa rates, while there were not statistically differences among the groups including cryoprotectants (trehalose+EDTA, GSSG, combined; 10.5±0.9%, 13.2±0.9%, 8.0±0.9%, respectively), at these groups fairly low rates (21.5±1.3%) were obtained in comparision to control, and differences between groups containing cryoprotectants and no cryoprotectants became important (P<0.05). Dead spermatoza with the lowest rate was attained at group with GSSG. At HOST, GSSG and combined groups gave the highest rates (40.5±2.7% and 27.0±1.4%, P<0.05). In conlusion, it was seen that cryoprotectants added to extender of Angora buck semen provided the significant contributions on some spermatological parameters after thawing.
