Morphological changes in dog' s sperm frozen with different diluents

- The aim of this research was to display the morphological changes in dog' s spermatozoa which were caused by freezing procedures using different diluents.

İn this experiment, semen was collected from dogs by manual stimulation. The percentage of abnormal sperm in fresh semen was 15.3 ± 6.0 %, acrosome, head, middle piece (spermatozoa with protoplasmic droplet and other middle piece defects) and tail defects (%) was averaged 0.5 ± 0.2, 0.1 ± 0.1, 10.3 ± 3.8 (3.4 ± 0.8 and 6.9 ± 3.0), 4.4 ± 2.3 respectively.

Ejaculates from dogs were diluted by Tris, Sodium citrate and Ringer. Postequilibration middle piece defects were established 14.4 ± 4.0 %, 13.3 ± 4.4 % and 15.2 ± 3.5 %. The percentage of spermatozoa with protoplasmic droplet and other middle piece defects (%) were recorded as 3. 7 ± 1.1 and 10. 7 ± 2.9, 3.9 ± 1.6 and 9.4 ± 2.8, 4.1 ± 1.1 and 11.0 ± 2.4 respectively.

Post-thawing middle piece defects were 16.2 ± 7.3, 14.7 ± 4.2 and 20.3 ± 6.9 respectively for frozen semen with the same diluents. The percentage of spermatozoa with protoplasmic droplet and other middle piece defets (%) were recorded as 2.9 ± 1.6 and 13.3 ± 5. 7, 1.8 ± 0.8 and 12.9 ± 3.4, 1.5 ± 0.9 and 18.8 ± 6.0 respectively.
