Freezing of Erbro Cock's Semen With Different Diluents and Cryoprotectants

- In this experiment, the effects of different semen diluents were assesed on freezing of semen and post-thawing spermatological characteristics via invitro datas in Erbro cocks.

In the study, 44 ejaculates collected from two cocks, three times in a week by massage method were treated with regard to principal spermatological characteristics. These are as follows: (1) Ejaculate volume averaged 0.5 ± 0.1 ml., (2) sperm motility 87.7 ± 0.9 %, (3) sperm concentrat1on 2.2 x 109 ± 0.1, pH 7.4 ± 0.1.

Mixed ejaculates were diluted with the two different semen diluents and frozen in liquid nitrogen vapour. Frozen thawed semen was assesed by means of thermoresistancetest (40 °C). Post-thawing motility and percentage of abnormal sperm vere 36.7 ± 2.6 % and 17.4 ± 2.1 % respectively for semen diluted with BPSV-1 containing 5 % Glycerol 5 % DMSO as cryoprotectants; 41.7 ± 2.1 and 21.7±2.8 with BPSV-2 containing 8 % Glycerol and 4 % DMSO. The same factures were recorded as 46.7 ± 1.9 %, 18.2 ± 1.1 % and 44.2 ± 1.9 %, 23.8 ± 2.9 % respectively for semen diluted with Ringer -1 and Ringer -2.

During the thermoresistance tests, it was observed that sperm motility ceased at 2. and 2.5 hours in semen diluted with BPSV-l and BPSV-2; at 4. hours with Ringer-l and Ringer-2; and at 6. hours in control group. Keywords :
