The Studies on In-Vitro Evaluation of Stallion Semen Frozen With Different Extenders

1 Hayvancılık Merkez Araştırma Enstitüsü Lalahan/ANKARA - The aim of this research was to investigate spermatolojical charecteristics of stallion semen frozen with lactose and tris extenders. This experiment was done in the Anadolu State Farm and five Pure Arabian Stallions were used.

Semen taken from five Arabian Stallions were extended in a way that to ensure 100x106 /cm3 motile spermatozoa by lactose and tris extenders and frozen in the macrotube in liquid nitrogen vapour.

In the semen after thawing of 50ºC-40 seconds the average values of semen; sperm motility (%),total percentage of abnormal sperm (%), percentage of abnormal acrosome (%) and thermo-rezistance test motility (%) in 1., 2. and 3. hours were recorded; %47.44; %31.73; %22.64; %22.74; %9.13 and %2.58 respectively for frozen semen by lactose.

In the semen after thawing of 50ºC-40 seconds the average values of sperm were recorded; %37.83; %36.06; %24.14; %17.58; %5.50 and %0.99 respectively as mentioned above for frozen semen by tris.

In the frozen semen by lactose extender, after thawing of 45ºC-30 seconds spermatolojical charecteristics were recorded; %42.33; %35.52; %24.59; %19.94; %6.74 and %1.13 respectively as mentioned above.

In the diluted semen by tris extender, after thawing of 45ºC-30 seconds spermatolojical charecteristics were recorded; %34.63; %38.39; %25.79; %15.94; %4.96 and %0.83 respectively as mentioned above. As a conclusion, extending stallion semen by lactose was more succesfull compare to by tris extenders and thawing at 50ºC-40 seconds was given better results than thawing at 45ºC-30 seconds. On the other hand, no relation was found semen extenders and thawing temperature and time. Keywords : Stallion, Semen, Dilution, Freezing, Thawing
