In vitro evaluation of saanen buck semen frozen in two different extenders

1 Ankara Üniversitesi Veteriner Fakültesi Dölerme ve Sun’i Tohumlama Anabilim Dalı- Ankara - The aim of this study was to evaluate for the effects of Tris-glucose-citric acid- egg yolk and Andromed extender, free of egg yolk, on sperm characteristics of buck semen after thawing in vitro. Two Saanen buck were used in this study. Semen was collected by an artificial vagina twice a week in autumn (mating season). After being divided into 2 equal aliquots, the collected semen diluted in tris diluent or andromed at 37ºC in water bath. Dilutions were carried out at a proportion of 1:10. The diluted semen was placed in 0.25 ml plastics straws and equilibrated (at 4ºC 2.5 hours) and frozen in vapour of liquid nitrogen (LN) (15 minutes at -110/-130 º C). Frozen straws were stored in liquid nitrogen (-196 ºC). Straws were thawed in the water bath at 37ºC for 30 seconds and spermatological examinations were carried out. The results demonstrated that spermatozoa motility (%), abnormal spermatozoa rate (%), acrosome abnormal rate (%) and hypo-osmotic swelling test (HOST) rate (%) were in frozen-thawed semen processed in Andromed (34±4.1, 45.40±3.7, 24.80±2.8, 36±3.5) were significantly (P<0.05) higher compare to Tris-egg yolk extender (20±3.5, 62.40±5.7, 42.80±2.7, 27.20±2.3). Regarding dead spermatozoa rate (%), there was no difference in between two extenders (51.80±6.3 Andromed versus 59.40±5.5 Tris; P>0.05). Consequently, it was found that Andromed was better than tris according to some sperm parameters evaluated postthaw. Keywords : Buck semen, extender, freezing, spermatological parameters
